A dramatic acceleration of the enantioselective copper-catalyzed conjugate reduction of alpha,beta-unsaturated lactones, lactams, and esters is reported upon addition of alcohol additives. Good to excellent yields and enantioselectivities were realized using a catalyst generated in situ from CuCl2·H2O, t-BuONa, p-tol-BINAP, and PMHS, and this methodology was applied to the synthesis of (-)-Paroxetine.

Blockage of more than one oncoprotein or pathway is now a standard approach in modern cancer therapy. Multiple inhibition is typically achieved with two or more drugs. Herein, we describe a pharmacophore merging strategy combining the JAK2/FLT3 inhibitor pacritnib with the pan-HDAC inhibitor, vorinostat, to create bispecific single molecules with both JAK and HDAC targeted inhibition. A preferred ether hydroxamate, 51, inhibits JAK2 and HDAC6 with low nanomolar potency, is <100 nM potent against HDACs 2 and 10, submicromolar potent against HDACs 1, 8, and 11, and >50-fold selective for JAK2 in a panel of 97 kinases. Broad cellular antiproliferative potency is supported by demonstration of JAK-STAT and HDAC pathway blockade in several hematological cell lines, inhibition of colony formation in HEL cells, and analysis of apoptosis. This study provides new tool compounds for further exploration of dual JAK-HDAC pathway inhibiton achieved with a single molecule.

The invention pertains to the use of Group 8 transition metal carbene complexes as catalysts for olefin cross-metathesis reactions. In particular, ruthenium and osmium alkylidene complexes substituted with an N-heterocyclic carbene ligand are used to catalyze cross-metathesis reactions to provide a variety of substituted and functionalized olefins, including phosphonate-substituted olefins, directly halogenated olefins, 1,1,2-trisubstituted olefins, and quaternary allylic olefins. The invention further provides a method for creating functional diversity using the aforementioned complexes to catalyze cross-metathesis reactions of a first olefinic reactant, which may or may not be substituted with a functional group, with each of a plurality of different olefinic reactants, which may or may not be substituted with functional groups, to give a plurality of structurally distinct olefinic products. The methodology of the invention is also useful in facilitating the stereoselective synthesis of 1,2-disubstituted olefins in the cis configuration.

Highly-functionalised difluorinated cyclooctenones were synthesised from trifluoroethanol using either metallated difluoroenol acetal or carbamate chemistry, followed by a [2,3]-Wittig rearrangement or aldol reaction. Efficient RCM reactions afforded the title compounds which showed rather restricted fluxional behaviour by VT 19F NMR. Topological characterisation by molecular modelling and NOESY/ROESY experiments offered a number of challenges, but allowed the identification of two favoured boat-chair conformers which interconverted by pseudorotation with relatively large activation barriers. The activity and stereoselectivity of phosphane- and N-heterocyclic carbene (NHC)-containing ruthenium benzylidene complexes have been evaluated in macrocyclic ring-closing olefin metathesis to produce unsaturated lactones and lactams. The success of the macrocyclization depends on the nature of the ligand (phosphane or N-heterocyclic carbene) on the ruthenium center and on the NHC properties. As for stereoselectivity, E/Z ratios seem to be influenced not only by the nature of the ruthenium catalyst but also by the thermodynamic stabilities of the resulting unsaturated macrocycles, as confirmed by theoretical results. The synthesis of 14-and 15-membered macrolactones and macrolactams by ruthenium-catalyzed ring-closing metathesis is investigated. The reaction outcome is influenced both by the nature of the catalyst and the diene substrate.

An efficient double bond migration-ring closing metathesis reaction leading to cycloheptene derivatives is observed when specific sterically congested 1,9-dienes are treated with the Grubbs’ imidazolidene ruthenium catalyst. The simultaneous use of the Grubbs’ catalyst and RuClH(CO)(PPh3) 3 facilitates the tandem bond migration-metathesis process. RuClH(CO)(PPh3)3 alone is capable of triggering an unactivated double bond migration that may have preparative applications.
