We have developed a simple and straightforward procedure for the enantioselective preparation of densely substituted bicyclic and tricyclic nitrogen heterocycles using conveniently substituted enantiopure pyrrolidines as common synthetic intermediates, which are easily accessible by our recently developed organocatalytic enantioselective [3+2] cycloaddition of alpha,beta-unsaturated aldehydes and azomethine ylides. The designed synthetic pathway makes use of a ring-closing metathesis reaction for building up the pyrrolizidine and indolizidine skeletons, while the access to the hexahydrocyclopenta[a]pyrrolizine structure has been carried out relying on a fully diastereoselective intramolecular Pauson-Khand reaction.

We report the first use of solution-phase halogen bonding to control and facilitate the assembly of an interlocked structure through the bromide anion-templated formation of a rotaxane based upon an iodotriazolium axle. Synthesis of novel pyrazole-fused heterocycles, i.e., dihydro-1H- or 2H-oxepino[3,2-c] pyrazoles (6 or 7) from 4-allyloxy-1H-pyrazoles (1) via combination of Claisen rearrangement and ring-closing metathesis (RCM) has been achieved. A suitable catalyst for the RCM of 5-allyl- 4-allyloxy-1H-pyrazoles (4) was proved to be the Grubbs second generation catalyst (Grubbs2nd) to give the predicted RCM product at room temperature in three hours. The same reactions of the regioisomer, 3-allyl-4-allyloxy-1H-pyrazoles (5), also proceeded to give the corresponding RCM products. On the other hand, microwave aided RCM at 140 C on both of 4 and 5 afforded mixtures of isomeric products with double bond rearrangement from normal RCM products in spite of remarkable reduction of the reaction time to 10 min.

An indolinooxazolidine tagged N-heterocyclic carbene Ru olefin metathesis catalyst was synthesized and the molecular structure of this new Ru complex was determined by single crystal X-ray diffraction. This complex is a homogeneous catalyst and can be recovered by controlling the polarity of the indolinooxazolidine tag. Under acidic conditions the indolinooxazolidine tag exists as an open protonated form and under basic conditions the tag is in a closed form. The distribution of this catalyst in a two-phase system can be controlled by simply changing the pH, making the recovery of this catalyst easily obtainable.

Two ruthenium hydride complexes commonly proposed as agents of unintended isomerization during olefin metathesis are examined for their activity in isomerization of estragole, a representative allylbenzene. Neither proves kinetically competent to account for the levels of isomerization observed during cross-metathesis of estragole by the second-generation Grubbs catalyst. A structure-activity analysis of selected ruthenium hydride complexes indicates that higher isomerization activity correlates with a more electrophilic metal center. It wasn’t me: Two Ru hydrides thought to trigger double-bond migration during olefin metathesis are examined for their isomerization activity. Neither can account for the high levels of undesired isomerization seen during self-metathesis of estragole, a model allylbenzene substrate.
