Metal-centered oxidations facilitate the removal of ruthenium-based olefin metathesis catalysts

Commercially available catalysts (SIMes)(PCy3)Cl 2Ru(CHPh) (2) and (SIMes)Cl2Ru(CH-o-O-i-PrC 6H4) (3) (SIMes = 1,3-dimesitylimidazolin-2-ylidene) were found to display reversible Ru oxidations via a series of electrochemical measurements. The redox processes enabled the catalysts to be switched between two different states of activity in ring opening metathesis polymerizations and ring closing metathesis reactions, primarily through changes in catalyst solubility. Moreover, treating a solution of 2 dissolved in C6H 6/CH2Cl2/[1-butyl-3-methylimidazolium][PF 6] (6:1:1.1 v/v/v) with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone was found to remove >99.9% of the catalyst, as determined by UV/vis spectroscopy. The methodology described herein establishes a new approach for controlling the activities displayed by commercially available olefin metathesis catalysts and for removing residual Ru species using redox-driven processes.

Isothiourea-catalysed chemo- and enantioselective [2,3]-sigmatropic rearrangements of N,N-diallyl allylic ammonium ylides

The isothiourea-catalysed chemo- and enantioselective [2,3]-sigmatropic rearrangement of N,N-diallyl allylic ammonium ylides is explored as a key part of a route to free functionalised alpha-amino esters and piperidines. The [2,3]-sigmatropic rearrangement proceeds with excellent diastereo- and enantiocontrol (>95:5 dr, up to 97% ee), with the resultant N,N-diallyl alpha-amino esters undergoing either mono- or bis-N-allyl deprotection. Bis-N-allyl deprotection leads to free alpha-amino esters, while the mono-deprotection strategy has been utilized in the synthesis of a target functionalised piperidine.

Total synthesis of platencin

(Chemical Equation Presented) The asymmetric total synthesis of the newly discovered and potent antibiotic platencin has been achieved. The approach makes use of an asymmetric Diels-Alder reaction, a gold(I)-catalyzed cyclization, and a homoallyl radical rearrangement to forge the polycyclic architecture of this intriguing target (see scheme, SEM=2-(trimethylsilyl)ethoxymethyl).

Testing the 1,1,3,3-tetramethyldisiloxane linker in olefin metathesis

Compounds 12-15, possessing two styrenes connected by a silicon linker [1,1,3,3 tetramethyl-di-siloxane], were synthesized, characterized and used as model compounds for the ring-closing metathesis (RCM) catalyzed by commercially available ruthenium catalysts 1, 2 and 3. The RCM reactions of 12 and 15 in the presence of catalysts 1 or 2 resulted exclusively in the formation of (E)-stilbenes. The RCM reactions of 13 and 14, compounds possessing alkoxide substituents in the ortho position to styrene functionality, were not observed in the presence of 2, presumably due to the formation of inactive Hoveyda type ruthenium complexes. The RCM of mixture of 12 and 15, with 2, was used for the detailed examination of the mechanism of metathesis reactions investigated in this work. They revealed that both inter- and intramolecular metathesis is possible, in this case, despite the use of siloxane linker.

Cross-metathesis approach to produce precursors of nylon 12 and nylon 13 from microalgae

A two-step synthesis for producing methyl 12-aminododecanoate and 13-aminotridecanoate, the precursors of nylon 12 and nylon 13, from methyl oleate is described. First, methyl 11-cyano-9-undecenoate or 12-cyano-9-dodecenoate were prepared by cross metathesis of methyl oleate with either allyl cyanide or homoallyl cyanide, respectively. Subsequently, all the unsaturation of the two intermediates was hydrogenated to deliver the final products. This method represents the first synthesis of nylon 12 and 13 precursors from methyl oleate, an ester of an abundant and renewable natural fatty acid present in vegetable oil or microalgae. It also represents the shortest synthesis of nylon precursors from fatty acids, and as demonstrated in this study, can be directly applied to crude fatty acid methyl ester extracts from microalgae.

Exploiting catalyst characteristics: A protocol for increasing diastereoselectivity in a double ring-closing metathesis reaction

A procedure for obtaining increased selectivity in a diastereoselective double ring-closing metathesis reaction is presented. The key to success was design of a substrate, which directed the reaction mechanism through a selective pathway and use of a suitable catalyst combination to achieve the desired balance between reactivity and selectivity.

Cross-metathesis of vinyl halides. scope and limitations of ruthenium-based catalysts

The phosphine-free second-generation Blechert/Hoveyda-Grubbs catalyst Ru(=C(H)C6H4-o-O-i- Pr)(H2IMes)Cl2 (H2IMes ) 4,5-dihydro-1,3-dimesitylimidazol-2-ylidene) and Piers catalyst [Ru(=CHPCy3)- (H2IMes)Cl2]BF 4 (Cy ) cyclo-C6H11) for olefin metathesis effected cross-metathesis (CM) of vinyl chloride and 1,2-dichloroethene with several unhindered terminal and internal alkenes in up to 95% yield (5 mol % catalyst). In most cases, 1,2-dichloroethene was more successful than vinyl chloride. Ring-opening CM of cyclooctene provided greater yields than CM: with vinyl chloride, 93% yield; with 1,2- dichloroethene, <95%. Other common Ru-based catalysts failed to effect CM under similar conditions, but instead underwent rapid decomposition. The dimeric ruthenium-monochloromethylidene complex [Ru(=CHCl)(H2IMes)Cl(mu-Cl)]2 was isolated as a thermally unstable intermediate. CM reactions with 1,2-dibromoethene afforded 22% CM product in the best case; halide exchange with the catalyst was significant. CM reactions involving vinyl fluoride typically led to <3 turnovers, but the dimeric intermediate [Ru(=dCHF)(H 2IMes)Cl(mu-Cl)]2 was sufficiently long-lived to be characterized by single-crystal X-ray diffraction. Ring-opening CM of cyclooctene with vinyl fluoride (55% yield) was more favorable than CM. Expanding the borononucleotide family: Synthesis of borono-analogues of dCMP, dGMP and dAMP

We previously reported the synthesis of a borononucleotide analogue of thymidine monophosphate and its association towards the formation of a new borono-linked dinucleotide. Here we describe the completion of the set of four 2?-deoxyborononucleotide analogues of natural nucleotide monophosphates, namely the previously unknown dCbn, dGbn and dAbn. These analogues were all prepared from the respective 5?-aldehydic nucleosides through a homologation/reduction sequence. The borononucleotides were subsequently obtained by either borylation (dCbn and dGbn) or cross-metathesis (CM) in the presence of the Hoveyda-Grubbs catalyst (dAbn). The reversible formation of the corresponding dinucleotides between these new analogues and uridine was studied by 1H NMR, and semi-empirical calculations were carried out to provide bond length and electrostatic information that assess the structural similarities existing between these bioisosteres and their natural counterparts.
