Reactions of chloro-bridged dimeric Ru(II) complexes [{Ru(eta6-arene)Cl2}2] with bridging ligands 1,4-dicyanamidobenzene, N,N?-dicyano 4-4?-diaminobiphenyl, 2, 5-dichloro-1,4-dicyanamidobenzene and 2,5-dimethyl-1,4-dicyanamidobenzene (referred hereafter as dcdH2, bpH2, ddcl and dmcd) in dicloromethane at room temperature gave binuclear complexes with the general formulation [{Ru(eta6-arene)Cl2}2(mu-L)]. However, reactions of these bridging ligands with the complexes [Ru(eta6-arene)(P)Cl2](eta6-arene = benzene or p-cymene; P = PPh3, PEt3 or MePPri2) in methanol, in presence of NH4PF6, gave cationic arene complexes [{Ru(eta6-arene)(P)Cl}2(L)]2+ (L = dcdH2, bpH2, dmcd or ddcl). The reaction products have been characterized by physico-chemical methods viz., elemental analyses, IR, 1H-, 13C-, 31P-NMR, electronic and FAB mass spectra. The complexes under study are highly stable at room temperature. However, their solutions in coordinating solvents like acetonitrile or dimthylsulfoxide undergo substitution reactions to give substitutional products with the formulation RuCl2(sol)4. It has been confirmed by single crystal X-ray diffraction studies.

The cross-metathesis of styrene with various vinylsilanes, H2C=C(H)SiR3, catalyzed by [Cl2(PCy3)2Ru=CHPh] (1) to give (E)-silylstyrene, (E)-Ph(H)C=C(H)SiR3, and ethylene is reported. The reaction proceeds even at room temperature and is highly selective. Very high conversions are observed when R = OEt, OSiMe3 (?95%, 6 h, 2 mol % of 1). The conversion significantly decreases with increasing substitution of Me for OR?. The metathesis is reversible. Therefore, removal of ethylene is critical for achieving high conversions. From the study of stoichiometric reactions of 1 with vinylsilanes it follows that in the series SiR3 = Si(OEt)3, SiMe(OEt)2, SiMe2OEt, SiMe3 and SiR3 = Si(OSiMe3)3, SiMe(OSiMe3)2, SiMe2-(OSiMe3), SiMe3 the conversion rate increases, but simultaneously the selectivity of the metathesis decreases. The decreasing selectivity readily accounts for the decreasing efficiency in the catalytic metathesis. The product distribution of reactions of styrene-d8 with H2C= C(H)SiR3 (R = OEt, OSiMe3) in the presence of 1 provides evidence for a metallacarbene mechanism involving [Ru]=CHPh and [Ru]=CH2 species.

A series of Ru(bpy)2-dioxolene complexes 1-4 (bpy = 2,2?-bipyridine) and corresponding Ru(dcb)2-dioxolene complexes 5-8 (dcbH2 = 2,2?-bipyridine-4,4?-dicarboxylic acid) have been prepared, and their spectroelectrochemical behavior in solution has been investigated. The complexes show reversible electrochemical behavior accompanied by a strong NIR absorption in their semiquinone forms due to a Ru(dpi) ? sq(pi*) MLCT band. Complete quenching of the NIR absorption occurs both upon oxidation (to the quinone form) and upon reduction (to the catechol form) very close to 0 V. The color of the systems can be tuned by using a wide range of ligands. The complexes 5-8 can be anchored onto nanocrystalline inorganic semiconductors allowing incorporation into potential electrochromic devices. As a proof of principle, compound 8 has been adsorbed on nanocrystalline Sb-doped SnO2 supported on FTO glass, and it displays reversibly switchable electrochromic behavior in the NIR.

Pinnatoxins belong to an emerging class of potent marine toxins of the cyclic imine group. Detailed studies of their biological effects have been impeded by unavailability of the complex natural product from natural sources. This work describes the development of a robust, scalable synthetic sequence relying on a convergent strategy that delivered a sufficient amount of the toxin for detailed biological studies and its commercialization for use by other research groups and regulatory agencies. A central transformation in the synthesis is the highly diastereoselective Ireland-Claisen rearrangement of a complex alpha,alpha-disubstituted allylic ester based on a unique mode for stereoselective enolization through a chirality match between the substrate and the lithium amide base. With synthetic pinnatoxin A, a detailed study has been performed that provides conclusive evidence for its mode of action as a potent inhibitor of nicotinic acetylcholine receptors selective for the human neuronal alpha7 subtype. The comprehensive electrophysiological, biochemical, and computational studies support the view that the spiroimine subunit of pinnatoxins is critical for blocking nicotinic acetylcholine receptor subtypes, as evidenced by analyzing the effect of a synthetic analogue of pinnatoxin A containing an open form of the imine ring. Our studies have paved the way for the production of certified standards to be used for mass-spectrometric determination of these toxins in marine matrices and for the development of tests to detect these toxins in contaminated shellfish.

Blockage of more than one oncoprotein or pathway is now a standard approach in modern cancer therapy. Multiple inhibition is typically achieved with two or more drugs. Herein, we describe a pharmacophore merging strategy combining the JAK2/FLT3 inhibitor pacritnib with the pan-HDAC inhibitor, vorinostat, to create bispecific single molecules with both JAK and HDAC targeted inhibition. A preferred ether hydroxamate, 51, inhibits JAK2 and HDAC6 with low nanomolar potency, is <100 nM potent against HDACs 2 and 10, submicromolar potent against HDACs 1, 8, and 11, and >50-fold selective for JAK2 in a panel of 97 kinases. Broad cellular antiproliferative potency is supported by demonstration of JAK-STAT and HDAC pathway blockade in several hematological cell lines, inhibition of colony formation in HEL cells, and analysis of apoptosis. This study provides new tool compounds for further exploration of dual JAK-HDAC pathway inhibiton achieved with a single molecule.

We performed measurements of magnetic susceptibility, electrical resistivity, and magnetoresistance in magnetic fields of up to 18 T in the magnetic superconductor RuGd1.4Ce0.6Sr2Cu2O10 – delta synthesized in oxygen pressures up to 95 atm. The magnetic-susceptibility data show the occurrence of an antiferromagnetic state below TN? 175 K, followed by the development of a weak ferromagnetic state near TM ? 100 K, and followed further by the onset of superconductivity (SC) at Tc ? 42 K. The electrical resistivity as a function of temperature shows an evolution from nonmetal-to-SC behavior in samples prepared in a flux of O2 to a well defined metal-like behavior in samples prepared under 95 atm pressure of O2. The electron-phonon coupling constant was calculated from transport data to be lambdatr?0.17, a value comparable with other cuprates, indicating weak electron-phonon coupling in these ruthenates. The values of the upper critical field Hc2 for the O2 high-pressure treated samples were obtained from the magnetoresistivity data yielding Hc2ab(0)?39 T, and the out-of-plane superconducting coherence length xic(0) ? (0) ? 28 A . Based on the similarities between these ruthenates and the superconductor YBa2Cu3O7 – delta, we estimated Hc2c(0) ? 8 T and xiab(0) ? 140 A . We used these parameters to discuss the coexistence of long-range magnetic order and superconductivity on a microscopic scale on these materials.
