A full account of the catalysis of hydrosilylation of the C=C bond in olefins, their derivatives with functional groups as well as in vinyl-trisubstituted silanes by ruthenium(II) and ruthenium(III) phosphine precursors is given.The ruthenium complexes are far more efficient catalysts for the hydrosilylation of 1-alkenes and vinyl-substituted silanes than for the substituted olefins and unsaturated esters.General features characterizing all hydrosilylation reactions catalyzed by the above catalysts are as follows: the reaction proceeds with alkoxy-substituted silanes(also with vinylsilanes) in the absence of solvent, and is enhanced (for RuII and olefins occurs exclusively) in the presence of molecular oxygen.Two general mechanisms are proposed for hydrosilylation of olefins and of vinylsilanes, respectively, which account for most of the experimental observations.

Rhodium- and iridium-mediated asymmetric hydrogenations of a number of olefins and one imine give products with only low to moderate enantiomeric excess, while in the ruthenium-catalyzed hydrogenation of ketones a maximum e.e. of 82% is obtained. The low enantioselectivities are assumed to be related to the conformational flexibility of bifep-type ligands.

We have established an efficient synthetic methodology for the 13-oxyingenol natural derivative (13-oxyingenol-13-dodecanoate-20-hexanoate), featuring a ring-closing olefin metathesis reaction for the ?direct? construction of a highly strained inside-outside framework and a Mislow-Evans-type [2,3]-sigmatropic rearrangement for the stereoselective introduction of the hydroxy group at C5. We also synthesized artificial analogs of 13-oxyingenol and ingenol by using our synthetic strategy. In vitro activation assays of protein kinase C (PKC) alpha and delta revealed that the dodecanoyl group at O13 on 13-oxyingenol analogs had a significant role in PKCdelta activation. The PKCalpha- or PKCdelta-activating 13-oxyingenol and ingenol analogs induced both distinct morphological changes and increases of CD11b expression in HL-60 cells, which would be typical signs of HL-60 cell differentiation to macrophage-like cells, as expected by previous reports. Intriguingly, however, similar differentiation phenotypes were observed with the use of 13-oxyingenol natural derivatives and 13-oxyingenol-13-dodecanoate showing a remarkably less potent PKCalpha or PKCdelta activation ability, which the PKC inhibitor Goe6983 diminished. This indicated the involvement of other PKC isozymes or related kinase activities. 13-Oxyingenol analogs, which induced HL-60 cell differentiation, also induced HL-60 cell death, similar to the action of a phorbol ester, a strong PKC activator.

The reaction is the first demonstration of direct sp3 C-H bond activation alpha to nitrogen followed by carbon-carbon bond formation under aerobic oxidation conditions. The catalytic oxidation seems to proceed by (i) alpha-C-H activation of tertiary amines by the ruthenium catalyst to give an iminium ion/ruthenium hydride intermediate, (ii) reaction with molecular oxygen to give an iminium ion/ruthenium hydroperoxide, (iii) reaction with HCN to give the alpha-aminonitrile product, H2O2, and Ru species, (iv) generation of oxoruthenium species from the reaction of Ru species with H2O2, and (v) reaction of oxoruthenium species with tertiary amines to give alpha-aminonitriles. On the basis of the last two pathways, a new type of ruthenium-catalyzed oxidative cyanation of tertiary amines with H2O2 to give alpha-aminonitriles was established. The alpha-aminonitriles thus obtained can be readily converted to alpha-amino acids, diamines, and various nitrogen-containing heterocyclic compounds.

The complexes RuCl3(CTZ)3·2CH3OH (1) and RuCl3(KTZ)2(H2O)·2H2O (2) were prepared by reaction of RuCl3·3H2O with CTZ and KTZ, respectively, while RuCl2(KTZ)2 (4) was prepared by reaction of RuCl2(CH3CN)4 with KTZ (CTZ = 1-[(2-chlorophenyl)diphenylmethyl-1H-imidazole, and KTZ = cis-1-acetyl-4-[4-[[2-(2,4-dichlorophenyl)-2-(1H-imidazol-1-ylmethyl)-1,3-dio xolan-4-yl]methoxy]phenyl]piperazine. All the complexes were characterized by NMR spectroscopy and for the paramagnetic species EPR spectroscopy was also employed. The new compounds were tested for in vitro activity against cultures of epimastigotes of Trypanosoma cruzi, the causative agent of Chagas disease, and compared with RuCl2(CTZ)2 (3) (reported previously) in order to establish some structure-activity correlations. At concentrations of 10-6 M (DMSO), all the complexes showed higher activity than the parental organic drug against the epimastigote form of the parasite, and Ru(II) complexes seem to be more active than their Ru(III) counterparts for a given nitrogen-donor ligand.
