Synthesis of Aromatic Retinoids and Curcuminoids and Evaluation of their Antiproliferative, Antiradical, and Anti-inflammatory Activities

Natural retinoids and curcuminoids are known for their broad spectrum of biological properties, such as antioxidant, anti-inflammatory, antitumor, and so forth. In this work, a convenient synthesis of aromatic retinoids and curcuminoids from vinyl or allyl ketones, and the corresponding alcohols, using olefin metathesis as a key reaction, was elaborated. The best yields and diastereoselectivities were obtained from allylic or homoallylic alcohols by employing the two-step cross-metathesis/oxidation procedure. The synthesized analogues were tested for their antiproliferative activity on human cancer cell lines of various origin (leukemia CEM, adenocarcinoma MCF7, cervical carcinoma HeLa) as well as for their antioxidant and anti-inflammatory activity in vitro. All examined derivatives exhibited strong anti-inflammatory activity in vitro without affecting cell viability. They also showed strong cytotoxicity against leukemia cell line CEM, except for 18 and 35. The antioxidant activity of the tested compounds was rather weak.

Sequence-Controlled Polymers Through Entropy-Driven Ring-Opening Metathesis Polymerization: Theory, Molecular Weight Control, and Monomer Design

The bulk properties of a copolymer are directly affected by monomer sequence, yet efficient, scalable, and controllable syntheses of sequenced copolymers remain a defining challenge in polymer science. We have previously demonstrated, using polymers prepared by a step-growth synthesis, that hydrolytic degradation of poly(lactic-co-glycolic acid)s is dramatically affected by sequence. While much was learned, the step-growth mechanism gave no molecular weight control, unpredictable yields, and meager scalability. Herein, we describe the synthesis of closely related sequenced polyesters prepared by entropy-driven ring-opening metathesis polymerization (ED-ROMP) of strainless macromonomers with imbedded monomer sequences of lactic, glycolic, 6-hydroxy hexanoic, and syringic acids. The incorporation of ethylene glycol and metathesis linkers facilitated synthesis and provided the olefin functionality needed for ED-ROMP. Ring-closing to prepare the cyclic macromonomers was demonstrated using both ring-closing metathesis and macrolactonization reactions. Polymerization produced macromolecules with controlled molecular weights on a multigram scale. To further enhance molecular weight control, the macromonomers were prepared with cis-olefins in the metathesis-active segment. Under these selectivity-enhanced (SEED-ROMP) conditions, first-order kinetics and narrow dispersities were observed and the effect of catalyst initiation rate on the polymerization was investigated. Enhanced living character was further demonstrated through the preparation of block copolymers. Computational analysis suggested that the enhanced polymerization kinetics were due to the cis-macrocyclic olefin being less flexible and having a larger population of metathesis-reactive conformers. Although used for polyesters in this investigation, SEED-ROMP represents a general method for incorporation of sequenced segments into molecular weight-controlled polymers.

Environmental sensitivity of Ru(II) complexes: The role of the accessory ligands

A suite of Ru(II) complexes in which one ligand is pH responsive and the other two are varied in an effort to achieve improved photophysics has been synthesized and their potential as pH reporters assessed. The more general purpose of the study was to examine the role of the accessory ligands in heteroleptic reporter complexes and the degree to which such ligands can affect the performance of luminescent reporters. For this suite of complexes, judicious choice of the accessory ligand can alter both the pKa* and the dynamic range of response. It was found that the emission color and brightness were influenced by pH, but the lifetimes were only weakly affected. Surprisingly, some accessory ligands which should have improved luminescent properties essentially turned off the pH response. Several possible reasons for this observation are explored. It is suggested, and density functional theory (DFT) calculations support, that the relative pi* levels of the pH sensitive and the accessory ligands are critical.

Olefin metathesis catalysts containing acyclic diaminocarbenes

The first examples of ruthenium-based olefin metathesis catalysts containing acyclic diaminocarbene (ADC) ligands are reported. Complexes of the type (ADC)(SIMeS)Cl2Ru=CHPh and (ADC)Cl2Ru=CH(2- isopropoxy)Ph (ADC = N,N’-dimethylformamidin-2-ylidene or N,N’-bis(2,6-di- isopropylphenyl)-N,N’-dimethylformamidin-2-ylidene; SIMes = 1,3- dimesitylimidazolin-2-ylidene) were synthesized and studied in solution as well as in the solid state. Depending on their N-substituents and the metal center to which they were coordinated, the aforementioned ADC ligands were found to adopt different conformations. Preliminary investigations revealed that these Ru complexes exhibited high catalytic activities in a variety of olefin metathesis reactions at elevated temperatures and afforded cross-metathesis products with significantly lower E:Z ratios than catalysts containing analogous N-heterocyclic carbene ligands.
